Theophylline Clearance of Korean population in comparison with American

Abstract
Theophylline (£±, £³£­dimethyl xanthine) is commonly used in patients with acute and chronic reversible air way diseases. Because of its narrow therapeutic range and side effects related with toxic theophylline blood level, therapeutic drug monitoring is essential to maintain safe and effective dosing. Especially, the determination of initial dose is extremely important during the acute phase of asthmatic attacks. The maintenance dose of initial infusion can be calculated from each patient¢¥s clearance value. Population theophylline clearance value of Americans were reporter by Jusko. Up to now, no clinical data on pharmacokinetic parameters (clearance, volume of distribution, etc) based on Korean population are available.
This fact makes it more difficult to keep patients in safe and effective therapeutic ranges of theophylline, when empirically dosed. Therefore, population theophylline clearance values of Koreans are vitally required for the safe dosing for asthma patients. This study was aimed to explore the theophylline clearance of Koreans by comparing with that of Americans reported by Jusko. As a result of this study, we could not deny the null hypothesis that theophylline clearance value of Americans is similar to that of Koreans. It is assumed that the clearance values of theophylline derived from Americans may by useful for dosage individualization of theophylline therapy in Korean asthma patients.
